Proteolysis targeting chimeras (PROTACs) have emerged as a novel biochemical modality that promotes ubiquitination and proteasomal degradation of specific intracellular protein targets. Recent pre-clinical and clinical trials have revealed a potential for therapeutic efficacy of PROTACs targeting various hormone receptors. This session will describe the mechanisms of PROTACs and present the evidence and limitations of their use to deplete androgen receptor in polycystic ovary syndrome and estrogen receptor in advanced breast cancer.
